The most correct way to measure the pH value of the fixative is to use a pH meter that can be calibrated at three points and has temperature compensation. Calibration should be carried out using three standard buffer solutions according to the instructions of the pH meter. The pH meter used to measure the fixative solution generally uses pH 4.0 (potassium hydrogen phthalate), pH 6.86 (mixed phosphate), pH 9.18 (sodium tetraborate), and three-purpose standard buffer (sold in finished products on the market). Just do the correction. The calibration process must be carried out in accordance with the instructions of the pH meter. After the calibration is completed, verify whether the calibration is in place.
Verification after calibration of the pH meter: After calibration of the pH meter, rinse it with distilled water, and then put it into the standard buffer for calibration at will, such as pH 6.86 (mixed phosphate) standard buffer. If it is placed in pH 4.0 (potassium hydrogen phthalate) standard buffer, the value displayed by the pH meter should be between 3.98 and 4.02. Correct operation when measuring the stationary solution: rinse the pH meter (glass bulb part) with distilled water and absorb all the water with absorbent paper. Insert the pH meter into the stationary solution to be measured, gently vibrate the pH meter (about 10 seconds), and let the pH meter stand. After the value on the pH meter is stable (not jumping), read the number. This number is the pH value of the fixative.

1. The concentration of phosphate in phosphate buffer cannot be too high or too low. It is recommended to use a concentration of 0.02 mol. The osmotic pressure of this concentration is similar to that of cell fluid and tissue fluid.
2. After mixing phosphate buffer solution with pH 7.2~7.4 and formalin and leaving it for 12 hours, the pH value will drop by about 0.3, and some will drop more. It is related to the formic acid in the raw material (formalin solution) It depends on the content.
